Exonuclease III (E.coli) catalyzes the stepwise removal of mononucleotides from 3?-hydroxyl termini of duplex DNA. The preferred substrates are dsDNA with blunt or recessed 3?-, termini although the enzyme also acts at nicks in dsDNA to produce single-strand gaps. dsDNA with 3?-overhang are resistant to cleavage; when overhangs length is ≥ 4 bases，dsDNA are able to fully prevent ExoIII cleavage process. Exonuclease III has also been reported to have RNase H, 3?-phosphatase and AP-endonuclease activities.

Mismatch Endonuclease I is a Mg2 dependent DNA endonuclease that specifically cleaves mismatched base pairs (TT GG and TG mismatches). Mismatch Endonuclease I cleaves the 3rd phosphodiester bond on the 5 side of the mismatched base in both strands leaving a 5 bp overhang. While Mismatch Endonuclease I prefers to cleave TT GG and TG DNA mismatches it will also readily cleave TI GI and GU DNA mismatches. Additionally Mismatch Endonuclease I has been shown to nick the thymine containing strand of TC DNA mismatches and cleave TG and TU (DNARNA) mismatches albeit to a lesser extent than DNADNA mismatches.

Exonuclease III (Exo III) is an exodeoxyribonuclease that digests one strand of duplex DNA from a blunt end, 5′ overhang or nick. The enzyme acts in a 3′→5′ direction producing stretches of single-stranded DNA (1, 2). Exo III is not active on 3′ overhangs of 4 or more bases in length and ssDNA. The enzyme has also RNase H, 3′-DNA phosphatase, and apurinic DNA endonuclease activities. Exo III digestions proceed at a uniform rate yielding predictable and reproducible results.
